1. BsaXI cleaves DNA substrates twice to excise its recognition site generating a 27 base-pair fragment with 3-base 3' overhangs.
2. Addition of greater than 2 units of BsaXI in a 16 hour incubation is not recommended due to DNA binding.
3. May exhibit star activity in NEBuffer r1.1, and NEBuffer r2.1.
4. For enzymes that cannot be heat-inactivated, we recommend using a column for cleanup (such as the Monarch® PCR & DNA Cleanup Kit), or running the reaction on an agarose gel and then extracting the DNA (we recommend Monarch Gel Extraction Kit), or performing a phenol/chloroform extraction.
5. Not sensitive to CpG, dcm, or dam methylation

1. How can this enzyme be inactivated?
2. Do I have to set-up digests with Time-Saver™ qualified enzymes for 5-15 minutes? Can I digest longer?
3. I tested your restriction enzyme on the substrate DNA recommended by NEB, and it appears to be active, however it does not digest my DNA. What could be the reason?
4. Is this enzyme sensitive to dam, dcm or mammalian CpG methylation?
5. Can you tell me more about the switch from BSA to Recombinant Albumin (rAlbumin) in NEBuffers?
