1. NarI is an isoschizomer of KasI. NarI produces a 2-base 5´ extension whereas KasI produces a 4-base 5´ extension.
2. Demonstrates marked site preferences. On pBR322, the NarI site at 548 bp is cut very slowly.
3. Under standard restriction digest conditions NarI requires two sites for efficient digestion. However, it is possible to obtain digestion of a single site if an excess of enzyme is used. For substrates containing a single NarI site we would recommend the use of 20 units of enzymeper 1 ug of DNA substrate. Alternatively, the neoschizomer SfoI can be used.SfoI does not demonstrate as strong a site preference as NarI does.
4. This enzyme has shown to have lower activity on some supercoiled plasmids, with more than 1 unit required to digest 1 μg plasmid DNA. For complete digestion of 1 μg of plasmid DNA please follow our recommended digestion protocol.
5. Based on the stability of the enzyme in the reaction, incubations longer than 1 hr will not result in improved digestion, unless additional enzyme is added. Please refer to Restriction endonuclease survival in a reaction for more information regarding this topic.
6. Requires two or more sites for cleavage. Please review the enzyme list and recommendations in the following table as well as the background information article.
7. Blocked by CpG methylation.

1. I tested your restriction enzyme on the substrate DNA recommended by NEB, and it appears to be active, however it does not digest my DNA. What could be the reason?
