NEBNext Ultra II FS DNAライブラリー調製キットを用いて、100 ngのヒトNA19240ゲノムDNA（Coriell Institute）から16種類のライブラリーを調製した。アダプターおよびプライマーは、IDT® for Illumina® –TruSeq® DNA UD Indexes（Illumina # 20022370）またはNEBNext Multiplex Oligos for Illumina（96 Unique Dual Index Primer Pairs）を使用した。PCRの4サイクル後、ライブラリーをAgilent® TapeStation® 4000で定量化した。A）それぞれのワークフローを用いて調製した8種類のライブラリーの平均ライブラリー収量は、NEBNext AdaptorおよびUnique Dual Index Primer Pairsを使用した場合、収量が60%高いことを示した。B) 16種類のライブラリーのTapeStation結果では、IDT for Illuminaのアダプターとプライマーを用いて調製したライブラリーでアダプターダイマー（オレンジ色の矢印で表示）の存在が認められるが、NEBNextのアダプターとプライマーを用いて調製したライブラリーでは認められず、収量も高い。

NEBNext Ultra II FS DNAライブラリー調製キット（図1）およびNEBNext 96 Unique Dual Index Primer Pairsを用いて調製した96種類のライブラリーを等モル濃度でプールし、Illumina NovaSeq 6000（2 x 150 bp）でシーケンスした。すべてのライブラリーの総リード数を合計し、各ライブラリーが寄与した総リード数に占める割合を決定した（ライブラリーあたりの期待割合=1.04%）。96種類のライブラリーはすべて効率的にクラスター化され、ほぼ期待される頻度で表れていた。

ヒトNA19240ゲノムDNA（Coriell Institute）を用いて、96種類のライブラリーを調製した。ライブラリーの調製には、NEBNext Ultra II FS DNA library prep kit、Covaris-sheared DNAとNEBNext Ultra II FS DNA library prep kit、バイサルファイト変換を組み合わせたNEBNext Ultra II FS DNA library prep kit、またはNEBNext Ultra II FS DNA library prep kitのいずれかを使用した。NEBNext 96 Unique Dual Index Primer Pairsを用いてライブラリーをPCR増幅し、ユニークなi5およびi7インデックスを含むライブラリーを生成した。ライブラリー収量を定量化し（Agilent® TapeStation® 4200）、96種類のライブラリーの全収量を合計し、各ライブラリーからの寄与を計算することで正規化した（ライブラリーあたりの期待割合 = 1.04%）。ライブラリー増幅効率は、各ライブラリー調製法で頑健であり、効率は96種類のユニークなインデックスプライマーの組み合わせすべてで均一であった。各バーは、少なくとも2回の実験結果の平均を示している。

NEBNext Ultra II DNA Library Prep Kit（NEB #E7645）およびNEBNext Multiplex Oligos for Illumina（96 Unique Dual Index Primer Pairs Set 1-4）を用いて調製した384種類のライブラリーを等モル濃度でプールし、Illumina NovaSeq® 6000でシーケンスした。すべてのライブラリーからの総リード数を合計し、各ライブラリーが総リード数に占める割合を決定した（ライブラリーあたりの期待割合 = 0.26%）。384のライブラリーはすべて効率的にクラスター化され、ほぼ期待される頻度で表れていた。
