RNA Inosine Endonuclease (RIE) is an RNA endonuclease that cleaves the 2nd phosphodiester backbone 3’ to a riboinosine (rI) base. The product leaves a nick in the RNA backbone with 3’-hydroxyl and 5’-phosphate. RNA Inosine Endonuclease has robust activity on single-stranded RNA, double-stranded RNA, and double-stranded RNA/DNA hybrids. RIE also has secondary minor activity cleaving deoxyinosine in single-stranded and double-stranded DNA.
