Mechanism of RNA Inosine Endonuclease

  • On RNA, catalyzes the cleavage of the 2nd phosphodiester backbone 3’ to a riboinosine (rI) base, leaving a 3’-hydroxyl and 5' phosphate terminus.
  • Robust activity on single-stranded RNA, double-stranded RNA, and double-stranded RNA/DNA hybrids.

  • RNA Inosine Endonuclease (RIE) is an RNA endonuclease that cleaves the 2nd phosphodiester backbone 3’ to a riboinosine (rI) base. The product leaves a nick in the RNA backbone with 3’-hydroxyl and 5’-phosphate. RNA Inosine Endonuclease has robust activity on single-stranded RNA, double-stranded RNA, and double-stranded RNA/DNA hybrids. RIE also has secondary minor activity cleaving deoxyinosine in single-stranded and double-stranded DNA.

    Source: An E. coli strain which contains the cloned Thermococcus kodakarensis Endonuclease V gene.

      One unit is defined as the amount of enzyme required to cleave 20 pmol of single stranded RNA oligonucleotide (FAM labeled 50 mer) containing a single internal riboinosine in a total reaction volume of 20 µL in 1X rCutSmart Buffer in 30 minutes at 65°C.

      50 mM NaCl
      0.1 mM EDTA
      0.2% Poloxamer 188
      50% Glycerol
      pH 7.4 @ 25°C

      Heat Inactivation

      90°C for 2 minutes

      1X rCutSmart Buffer containing 20 pmol of fluorescently labeled oligonucleotide in a total reaction volume of 20 µl for 30 minutes at 65 °C.
