TnX® Transposase (Unloaded) is an engineered transposase that, when loaded with mosaic-end adaptors, simultaneously fragments double-stranded DNA and incorporates adaptor sequences in a single-step reaction known as tagmentation. The unloaded format enables generation of custom transposomes for molecular biology workflows.

TnX Transposase (Unloaded) is assembled with customizable mosaic-end adaptor oligonucleotides and acts on double-stranded DNA to generate DNA fragments and incorporate adaptor sequences in a single enzymatic step (tagmentation). Mosaic-end adaptors are required but not included with TnX Transposase (Unloaded) (NEB #M2623).
TnX Transposase (Unloaded) was loaded with mosaic-end adaptor oligos then incubated with 50 ng of genomic DNA at 60°C for 10 minutes. The tagmentation reaction was stopped with 0.75% SDS at 60°C for 10 minutes. Fragmented molecules were visualized on High Sensitivity D5000 TapeStation® after 2X bead cleanup.

Unit Definition
One unit is defined as the amount of loaded enzyme required to fragment and add adaptors onto 50 ng of gDNA in 10 minutes at 60°C. Fragmented and tagged molecules are measured to be an average size of 350-450 base pairs.
