    • EnClose TelA Protelomerase is a telomere resolvase that cuts dsDNA at a 50 bp recognition sequence and leaves covalently closed ends at the site of cleavage through hairpin formation. A minimal 26 bp sequence can also be utilized by the enzyme while retaining full activity.

      An E. coli strain that carries a recombinant plasmid encoding an engineered TelA Protelomerase from the Gram-negative bacteria Agrobacterium tumefaciens and retains an N-terminal 8x His-tag.

        Unit Definition

        One unit is defined as the amount of enzyme required to cleave 0.5 µg of pMiniT2.0-TelA AhdI-linearized control plasmid (313 fmol of TelA recognition sites) in a total reaction volume of 50 µl in 30 minutes at 42°C in 1X Protelomerase Reaction Buffer.

        Incubate at 42°C

        20 mM Tris-HCl
        10 mM (NH4)2SO4
        10 mM KCl
        2 mM MgSO4
        0.001% Tween® 20
        (pH 8.8 @ 25°C)

        Heat Inactivation

        80°C for 10 minutes
