NEBNext® High Input Poly(A) mRNA Isolation Module

NEBNext High-Input Poly(A) mRNA Magnetic Isolation Moduleは、オリゴd(T)25結合常磁性ビーズを用いて、抽出済の大量のトータルRNA（0.2 mlの反応液量当たり5～50 µg）からインタクトなポリ(A)+ RNAを精製するよう設計されている。インタクトなmRNAは、約1時間で精製でき、少量で溶出できる。

  • NEBNext High-Input Poly(A) mRNA Magnetic Isolation Moduleは、抽出済の大量のトータルRNA（1反応あたり5～50 µg）からインタクトなポリ(A)+ RNAを分離できる。この技術は、オリゴd(T)25と常磁性ビーズの結合に基づいており、ポリ(A)+ RNAの高純度の精製に使用されている。本製品により、複数のサンプルを手動で処理することができるほか、自動ハイスループットアプリケーションに適合させることもできる。さらに、磁気分離技術により、少量でインタクトなmRNAを溶出できるため、溶出後のポリ(A)+ mRNAの沈殿は不要である。インタクトなポリ(A)+ RNAは、元のサンプル中のmRNA集団を反映しており、約1時間で採取できる。

    5 µg未満のインプットRNAについては、NEBNext® Poly(A) mRNA Magnetic Isolation Module（NEB #E7490）が推奨される。

    E3370ワイドレンジ

    A. External RNA Controls Consortium (ERCC) RNA及びSpike-In RNA Variants（SIRV Set 4, Lexogen®）を含む、50 μgまたは5 μgの総Universal Human Reference (UHR) RNA（Agilent®）からNEBNext High Input Poly(A) mRNA Isolation moduleを用いて精製したポリ(A)RNA。総収量は、Qubit® RNA High Sensitivity Assay Kit（Invitrogen®）を用いて評価した。ポリ(A)RNAは通常、トータルRNAの約1～5%である。 
    B. Total UHR RNA及び50 μgまたは5 μgのトータルRNAインプットからのポリ(A)精製RNAのBioanalyzer®（Agilent）の結果。 
    C. 転写産物の存在量は、NEBNext Ultra™ IIディレクショナルRNAライブラリー調製キットを用いて、40 ng（50 μgインプット）または8 ng（5 μgインプット）のポリ(A)精製RNAから調製したライブラリーから決定し、Illumina® NextSeq® 550シーケンサーでシーケンスした。各ライブラリーから600万のリードをサンプリングした。

    E3370の保持

    ポリ(A)RNAは、Dynabeads® mRNA Purification Kit（Invitrogen）、Poly(A)Purist™ MAG（Invitrogen）またはNEBNext High Input Poly(A) mRNA Magnetic Isolation Moduleを用いて、50 μg Universal Human Reference RNA（UHR、Agilent）またはMonarch® Total RNA Miniprep Kitを用いてマウス腎臓組織、もしくはS. cerevisiae（酵母）から抽出したRNAから精製した。 
    A. 全体及び個々のポリ(A)精製RNAのBioanalyzerの泳動結果。 
    B. 3回（UHR及びマウスポリ(A)サンプル）または2回（トータルRNA及び酵母ポリ(A)RNAサンプル）のシーケンシングから、全体及び個々のポリ(A)精製RNAサンプルのrRNAの割合を標準偏差と共に測定した。ライブラリーは、NEBNext Ultra™ II Directional RNA Library Prep Kitを用いて40 ngポリ(A)精製RNAから調製し、Illumina NextSeq 550でシーケンスした。各ライブラリーから600万のリードをサンプリングした。

    E3370高収量 
    記載された方法を用いて精製した、ポリ(A)精製済のUniversal Human Reference（UHR）RNA 400 ngを、GridION®シーケンサー（Oxford Nanopore Technologies®）でDirect RNA Sequencing（ONT #SQK-RNA002）用に調製した。記載された方法を用いて精製したポリ(A)精製済のUHR RNA 100 ngを、GridIONシーケンサーでDirect cDNAシーケンシング（ONT #SQK-DCS109）用に調製した。  
    A. ライブラリーの収量は、Qubit dsDNA High Sensitivity Assay Kit（Invitrogen）を用いて評価した。複製の平均値を標準偏差と共に示す。 
    B. 複製Direct RNAシーケンシング及びDirect cDNAシーケンシングから得られたリードの平均マッピング率並びに標準偏差。

    NEBNext High Input Poly(A) mRNA Isolation Moduleを用いてUHR RNAからポリ(A)精製し、調製したライブラリーを用いてOxford Nanopore Technologiesプラットフォーム上で実行した、代表的なDirect RNA（A）及びDirect cDNA（B）シーケンシングから得られた上位1000転写物の5´➝3´転写物のカバレッジ。
