Nickel Purification (His-tag)

Poly-histidine tagging is widely employed for the purification of recombinant target proteins via immobilized metal affinity chromatography (IMAC). The advantages include small tag size and high affinity and specificity of poly-His tag binding to divalent metals at neutral pH. To enable rapid purification of His-tagged proteins, NEB offers NEBExpress Ni-NTA Magnetic Beads (NEB #S1423), NEBExpress Nickel Spin Columns (NEB #S1427) and NEBExpress Nickel Resin (NEB #S1428). NEBExpress Ni-NTA Magnetic Beads enable the small-scale isolation and purification of His-tagged fusion proteins in manual or automated formats. They are prepared with agarose-based, super-paramagnetic microparticles that provide high binding capacity and fast magnetic response. NEBExpress Ni Resin is recommended for the isolation and purification of His-tagged fusion proteins and is intended for use in gravity or pressure flow columns and batch purifications. NEBExpress Nickel Spin Columns are pre-packed with agarose-based microparticles ranging in size from 10–100 µm for the small-scale isolation and purification of His-tagged fusion proteins.
