RNA Markers and Ladders
NEB offers several RNA markers and ladders as gel electrophoresis sizing standards for single-stranded and double-stranded RNA, featuring double intensity reference bands and recommendations for RNA species, gel types, handling, and staining.
RNA gel electrophoresis is a fundamental technique to reveal RNA quality, relative abundance, and sizing. RNA gels are useful for identifying the presence of RNA species, validating RNA extraction or RNA synthesis results, and isolating specific RNA fragments for downstream analysis. Common downstream techniques include RT-PCR, RT-qPCR, RNA-seq and Northern Blot. RNA electrophoresis can be performed with denaturing polyacrylamide-urea gels, denaturing agarose gels and native agarose gels.
- The ssRNA Ladder (NEB #N0362S) has a size range of 500 bases - 9,000 bases and is compatible with denaturing and native gels.
- The dsRNA Ladder (NEB #N0363S) has a size range of 21 - 500 base pairs, featuring a double intensity 80 base pair reference band. It is suitable for use in both non-denaturing polyacrylamide and agarose gels.
- The Low Range ssRNA Ladder (NEB #N0364) has a size range of 50 bases - 1,000 bases, featuring a double intensity fragment to serve as a user-friendly reference band. It is supplied with 2X sample buffer and is compatible with denaturing polyacrylamide-urea gels, denaturing agarose gels and native agarose gels.
- The microRNA Marker (NEB #N2102) for miRNA is a set of 17, 21 and 25 residue marker oligos in a ready-to-load denaturing solution, ideally used as a size marker on denaturing polyacrylamide gels or Northern blots and best visualized with SYBR-Gold. It is supplied with a 3´-biotinylated 21-mer oligonucleotide probe that can be labeled with γ32-P-ATP using T4 PNK (NEB #M0201).
- RNA Loading Dye (2X) (NEB #B0363) is pre-mixed for use with denaturing and non-denaturing gels.

- What types of gel and staining dyes can I use for NEB’s Low Range ssRNA Ladder (N0364S)?
- Why the bands of the Low Range ssRNA Ladder on my 2% agarose-TBE gel are not resolved well?
- Why do I have extra bands of the Low Range ssRNA Ladder on my denaturing 6% TBE-Urea gel?
- Will the Low Range ssRNA Ladder run on 10% TBE-Urea and 15% TBE-Urea gels?
- What is the concentration of RNA in the ssRNA Ladder (NEB #N0362) and Low Range ssRNA Ladder (NEB #N0364)?
- What types of gel and staining dyes can I use for NEB’s dsRNA Ladder (N0363S)?
- Can I use the microRNA Marker on a non-denaturing gel?
- Should I denature the microRNA Marker before loading on my gel?
- What is the difference between the siRNA marker and the microRNA marker (discontinued)?
- Can I use EtBr for staining the microRNA Marker after gel electrophoresis?
- Should I load more of the microRNA Marker so I can get better visibility of the bands after staining the gel with EtBr?
