Quick Tips – Freezing cells after sorting in the NEBNEXT® Single Cell/Low Input RNA library prep workflow

Learn how to best preserve RNA in your samples when freezing cells in the NEBNext Single Cell/Low Input RNA library prep workflow.
Script
When working with the NEBNext Single Cell/Low Input RNA Library Prep Kit or Module, freezing cells after cell sorting can be a little tricky.
To keep RNA intact, it’s best to sort the cells directly into the NEBNext lysis buffer and murine RNase inhibitor provided with the kit.
Then quickly spin the cells and freeze as soon as possible in an ethanol dry ice bath to prevent degradation of the RNA.
Cells stored this way are stable for several months. And of course, all the samples for any given project should be treated with the same workflow.
