FAQ: Do the tracking dyes present in Q5 Quick-Load™ interfere with downstream applications?
Q5 Quick-Load products contain two distinct dyes (yellow and blue) which are compatible with downstream applications such as DNA sequencing, ligation, and restriction digestion. For applications that require analysis of PCR products by absorbance or fluorescence, we recommend using colorless versions of the products (NEB #M0492 and NEB #M0494) or to purify the PCR products first. The Monarch PCR & DNA Cleanup Kit (NEB #T1130) is recommended for this task as it removes the dye while leaving the DNA intact for subsequent analysis. The Exo-CIP™ Rapid PCR Cleanup Kit (NEB #E1050) is also recommended to efficiently clean up PCR reactions.
