Double Digest Protocol using Two RE-Mix® Enzymes

Protocol

  1. Dilute up to 1 μg DNA to 36 μl with dH20
  2. Add 2 μl of each 10X RE-Mix
  3. Incubate for 15 minutes at 37°C
  4. Analyze by agarose gel electrophoresis

    Notes: Alternatively, double digest can be carried out in a 20 μl volume using 1 μl of each RE-Mix and a minimum incubation time of 30 minutes.
There is also a protocol for double digest with a standard restriction enzyme with RE-Mix.
