In general, library preparation of fragmented DNA, for use with the Ion Torrent™ sequencing platform, involves repair of 3′ and 5′ ends before ligation to an adaptor. After ligation, adaptor-DNA constructs are purified and size selected, and amplified via polymerase chain reaction (PCR). Libraries prepared in this manner are suitable for template preparation, the next step in the Ion Torrent sequencing workflow.
NEBNext® reagents are available for each step in the DNA library preparation workflow for the Ion Torrent platform. For DNA library preparation, reagents from NEB are available with or without reagents for enzyme-based fragmentation. Each of these reagents has been functionally validated by preparation of a genomic DNA library followed by Ion Torrent sequencing.
