FAQ: How do I use Thymine-DNA Glycosylase (TDG) (NEB #M0766) with the NEBNext® FFPE DNA Repair Mix?

The addition of TDG to the FFPE DNA Repair Mix will further decrease C to T artifacts, especially in CpG contexts. TDG is added during step 1.2.1 of the FFPE DNA Repair Mix protocol directly to the reaction mixture: after adding the 3 µL of the NEBNext® Ultra II End Prep Enzyme Mix, add 4 µL of TDG and follow the rest of the protocol as usual.
