FAQ: Does the EnClose™ Cell-free dbDNA™ Synthesis Kit (NEB #E9301) work with other buffers?

Although the enzymes in this kit work in other reaction buffers separately, it is important to note that the dbDNA Reaction Buffer has been optimized to enable the entire enzymatic workflow from plasmid to dbDNA to occur in a single-pot reaction. Other buffers can be used but will not provide optimal dbDNA yields and additional intermediate cleanup steps may be required.
