FAQ: Can I dilute the RCA reaction before the EnClose™ TelN Protelomerase reaction step in the EnClose™ Cell-free dbDNA™ Synthesis protocol (NEB #E9301)?
We do not recommend diluting the RCA reaction before EnClose TelN Protelomerase is added in the workflow. Dilution of the reaction makes EnClose TelN Protelomerase less efficient and will decrease the yield of the desired dbDNA product. Because the completed RCA reaction is likely viscous, we recommend using a wide-bore pipette tip to achieve optimal mixing at this step.
