For the 96-reaction size (NEB #E3404), each well of the supplied 96-well primer plate contains a unique pair of one i5 index primer and one i7 index primer, therefore providing 96 unique dual indices. For the 24-reaction size, (NEB #E3392S) indices are provided in 24 wells of a 96-well plate, with each well containing a unique pair of one i5 index primer and one i7 index primer. There is no overlap between i5 and i7 index pairs provided in E3392 and E3404.
