Yes, the Monarch Spin gDNA Extraction Kit can be used to clean up phenol/chloroform purified gDNA by following the protocol for Genomic DNA Cleanup. However, recovery may be lower than the usual 80% because phenol/chloroform purified DNA typically yields longer fragments. Although the use of preheated elution buffer in the Monarch protocol facilitates the elution of large gDNA fragments, the fraction of gDNA that is longer than 80 kb will be eluted less efficiently from the silica matrix.
