FAQ: Can glycosidases be used in combination for extensive digestion?
Yes, combinations of glycosidases can be used in the same reaction mixture, for example the Protein Deglycosylation Mix II (NEB #P6044) is a combination of five glycosidases. Other combinations are possible, as long as the pH range allows a common reaction buffer. When exoglycosidases are added to a mix with endoglycosidases, it is possible that the released glycans will be trimmed by them, and therefore will not be recovered intact. Also, since pH changes might affect performance, more units or longer incubation time might be necessary when a given enzyme is acting in a non-typical buffer.
