FAQ: Can the RCA step in the EnClose™ Cell-free dbDNA™ Synthesis Kit (NEB #E9301) protocol be performed at a different temperature?
The phi29-XT DNA Polymerase used for the RCA reaction has an optimal temperature range of 40°C − 42°C. Amplification at temperatures below 37°C is not recommended.
