RNA structure profiling and nucleotide modification mapping at single-cell resolution
Structural features and nucleotide modifications in mRNAs play critical roles in gene regulation, influencing processes such as RNA decay and protein translation. Additionally, many noncoding RNAs—such as tRNAs and snRNAs—depend on distinct structures and modifications to carry out specific cellular functions. Although their regulatory importance is well established, high-throughput analysis of these features at the single-cell level remains challenging. The Kim lab utilizes novel NEB enzymes within single-cell sequencing platforms to map RNA structures and nucleotide modifications across the transcriptome of individual cells. Through these efforts, we aim to uncover how RNA features contribute to gene regulation across diverse cell types.
