A gel-like pellet may be visible in the column after the first elution. This contains RNA of interest that is not fully resuspended in the nuclease-free water. It is recommended to perform additional elution steps. A second elution (or third elution in the case of RNA sizes ≥ 9 kb) will recover remaining RNA of interest.
