FAQ: My sequence of interest (SOI) contains an XbaI recognition site. How can I remove the plasmid backbone without cutting my dbDNA product with XbaI during the enzymatic cleanup step?

If you cannot domesticate your sequence of interest to remove an XbaI site, you may use one of the alternative processing restriction enzymes optimized in the dbDNA vector backbone sequence: ApaLI, AseI, BamHI-HF, ClaI, FspI, NdeI, PacI, RsrII, SbfI-HF. Refer to the table below:
