FAQ: When using the Monarch Spin High-Capacity RNA Cleanup Kit (3 mg) (NEB #T2060), what if my starting sample volume is > 800 µl before adding Buffer BX and ethanol?

For samples with starting sample volume > 800 µl, it is recommended to partition the sample into aliquots and spilt the input across several columns. A starting sample volume of 800 µl will result in a final volume of 4.8 ml to be loaded onto the column (800 µl sample + 1600 µl Monarch Buffer BX + 2400 µl ethanol), which results in 8 column loading steps.

It is not recommended to process large volumes of starting sample > 800 µl, as this leads to an increase in the number of times a column is re-loaded, and may result in problems during elution. The Monarch Spin High-Capacity RNA Cleanup Kit (3 mg) is recommended for samples with higher starting concentration of RNA (e.g., 5–10 µg/µl or greater).
