FAQ: When using the Monarch Spin High-Capacity RNA Cleanup Kit (3 mg) (NEB #T2060), what happens if I use the wrong centrifugation speed during sample loading?

The recommended centrifugation speed during the sample loading steps for the Monarch Spin High-Capacity RNA Cleanup Kit (3 mg) is 10,000 x g.

If the sample loading spins were carried out at 16,000 x g instead, there is a possibility that a portion of the sample may be in the flow-through and collected in the collection tube. In this event, a pellet will be visible in the collection tube and can be recovered. Carefully discard the supernatant in the collection tube and resuspend the pellet with nuclease-free water. The resuspended sample can then be put through the RNA cleanup protocol once again and successfully recovered.
