Dnmt1 prefers to methylate cytosine residues in hemimethylated DNA at 5'...CG...3'. For DNA modification and protection application, M. SssI is preferred, as both unmethylated and hemimethylated DNA substrate are methylated efficiently by M. SssI (NEB# M0226).
