Here are some important things to keep in mind when preparing the NEBNext® Ultra II FS DNA reaction buffer and enzyme mix for use.

Some of our customers have asked how to prepare the Ultra II FS DNA reaction buffer and enzyme mix. First, thaw the Ultra II FS buffer and enzyme on ice. When they are thawed, do a quick spin. Break up any precipitate by pipetting up and down with a p200 pipette tip, until the precipitate is broken up. Proceed to vortex for 5 seconds and do a quick spin, then put the buffer back on ice until your ready to use it. Some flakes will remain undissolved, which is not a concern, we don’t see any difference in fragmentation due to a small amount of precipitate remaining. To prepare the enzyme mix, remove the enzyme vial from the ice, vortex it for 5 seconds and do a quick spin. Vortexing is necessary for consistency, and not harmful to the enzyme.
