Adaptor or Vector Ligation of cDNA Library (E6100)
Protocol
- Mix the following components in a sterile 1.5 ml microcentrifuge tube:
Purified, dA-Tailed cDNA: 23 μl
2X Quick Ligation Reaction Buffer: 25 μl
DNA Adaptors (15 μM) or vector: 1 μl
Quick T4 DNA Ligase: 1 μl

Total volume: 50 μl - Incubate 15 minutes at room temperature.
- Purify Adaptor Ligated cDNA using a PCR column purification kit, purifying the sample on one column and eluting in 10 μl sterile water or elution buffer.
