FAQ: Are there differences in ligation efficiency for T4 Rnl2tr and T4 Rnl2tr K227Q?
In NEB tests, ligations using T4 Rnl2tr and T4 Rnl2tr K227Q on identical substrates show that ligation efficiency reaches the same endpoints. However, T4 Rnl2tr K227 may require longer incubation times to reach the reaction endpoint. We recommend that pre-adenylated DNA linker ligations be incubated overnight at 16°C.
