Standard Protocol for Msz Exonuclease I Reaction (NEB #M0527)

1. Set up a 20 µl reaction as follows:

   COMPONENTS	20 µL REACTION
   ssDNA	up to 1 µg
   rCutSmart™ Buffer (10X) (NEB# 6004S)	2 µl
   Msz Exonuclease  I (NEB #M0527)	1 µl
   Nuclease free H2O (NEB# B1500S)*	up to 20 µl*

   *Scale larger reaction volumes proportionally
   
   
2. Incubate at 55°C for 15 minutes 
   
   
3. Stop reaction by heat-inactivation at 80°C for 1 minute or by chelation of Mg²⁺

Note: Most PCR buffers are compatible. No additional reaction buffer is necessary if enzyme is added to a PCR reaction.
