Use of the 3X Reducing Blue Protein Loading Dye for Protein Gel Samples (NEB #B7703)

  1. Prepare fresh 3X Reducing Blue Protein Loading Dye by adding 1/10 volume 30X Reducing Agent to 1 volume of 3X Blue Loading Dye. Please ensure that the 3X Reducing Blue Protein Loading Dye is completely thawed and mixed before use.

  2. Prepare samples by adding 1/2 volume of 3X Reducing Blue Protein Loading Dye (from step 1).

  3. Heat samples to 95–100°C for 3–5 minutes.

  4. After a quick microcentrifuge spin, load directly on to a gel. To ensure uniform mobility, load an equal volume of 1X Reducing Blue Protein Loading Dye into any unused wells.

  1.  The 3X Reducing Blue Protein Loading Dye, containing the reducing agent, should be stored at -20°C
