FAQ: How should I mix or resuspend Immobilized T4 DNA Ligase (NEB #M0569) beads prior to use?
Immobilized T4 DNA Ligase should be removed from -20°C and placed directly on ice before dispensing. Prior to any dispensing event, we recommend briefly spinning the tube in a mini-centrifuge, followed by mixing with a pipette to fully resuspend the beads and achieve homogeneity. Care must be taken as the solution is viscous: submerge only the tip of pipette when aspirating and do not create foam when pipette mixing. Pipette mixing can be performed by setting the volume of your pipette to ~50% of the volume in the tube (to avoid drawing air) and pipetting up and down at least 10 times. Once you have dispensed the intended amount, briefly spin down the tube in a mini centrifuge before returning the product to -20°C for storage.
