There are several factors to consider. In general, the substrate is prepared in batch and then subdivided for enzyme digestion; therefore the number of final exoglycosidase reactions desired needs to be determined. Furthermore, the sensitivity of the particular fluorescent label used determines the quantity of labelled glycan required for adequate signal detection. Some instant labels, such as Rapifluor, have a sensitive fluorescent detection and good signal intensity for mass detection and so less starting material is required.
