FAQ: Can Neuraminidase be used together in a digest with PNGase F and O-Glycosidase?
Yes. Conditions have been determined (FAQ#1) that allow O-glycosidase to be used under the same denaturing conditions used for PNGase F. PNGase F and O-glycosidase can be used concomitantly in a reaction with 1X G7 Reaction Buffer, 10% NP-40, Neuraminidase, H20 and your glycoprotein. Neuraminidase has a very broad pH range and is active between pH 4.5-8.5. It works very well on a protein that has been denatured, and Neuraminidase shows normal activity even in the presence of 1% BME, and 0.5% SDS.
