FAQ: Is there any difference in the methylation sensitivity between SacI-HF and SacI?
Yes, SacI-HF and SacI differ in their sensitivity to overlapping mammalian CpG methylation. SacI-HF™ is blocked by overlapping CpG methylation on both strands and impaired if only one strand is methylated. SacI is not blocked by any methylation. For more specific information regarding methylation, follow the link to the Restriction Enzyme database REBASE.
