FAQ: Will Thermolabile Exonuclease I digest my double-stranded DNA while removing ssDNA in the reaction?

Thermolabile Exonuclease I will not cleave dsDNA in most of PCR buffers (such as Q5 PCR buffer, Phusion PCR buffer, Thermopol buffer, Taq buffer, oneTaq quick-load buffer or NEBuffer 1-4). However, some PCR additive (such as in NEB# M0489 and Q5 high GC enhancer) will cause Thermolabile Exonuclease I to partially digest dsDNA. If this occurred, it is recommended to add an equal amount of water to the PCR sample before enzyme digestion.
