Quick Tips - Preventing media carryover in cell sorting for the NEBNEXT® Single Cell/Low Input RNA Kit

Learn how to prevent media carryover in cell sorting if you are unable to sort directly into the NEBNext Cell Lysis Buffer with murine RNase inhibitor.

When working with the NEBNext Single Cell/Low Input RNA Library Prep Kit or Module, it’s best to sort cells directly into NEBNext lysis buffer and murine RNase inhibitor. But if you cannot do that, you need to prevent media carryover, because media carryover will inhibit enzymatic activity. Wash the cells and resuspend them in PBS prior to sorting. Limit PBS carry over to no more than 5 ul. Ideally, you should sort into 2-3 ul of PBS, sort cells quickly and flash freeze, or use immediately.
