Site Directed Mutagenesis

Site directed mutagenesis describes changes made to a sequence in a targeted manner. The technique requires that a primer be designed that is complimentary to the template DNA, except for the mutation; it is therefore essential that the sequence of the template DNA be known. The annealed primer is then copied by DNA polymerase, which dutifully incorporates the mutation.
