EcoGII Methyltransferase Protocol (M0603)

1. Set up the following reaction on ice.
 COMPONENT  50 µl REACTION
 Nuclease-free Water  to 50 µl
CutSmart Buffer (10X)  5 µl (1X)
 S-adenosylmethionine*
  5 µl (160 µM)
 DNA  up to 1 µg
 Methyltransferase
  1 µl (5 units)
*Dilute SAM to 1.6 mM using the supplied 32 mM stock.

2. Incubate at 37°C for 60 minutes.

The amount of methylation can be optimized by varying amount of substrate, amount of enzyme and/or incubation time.
