
DNA Methylation Analysis
DNA methylation plays a pivotal role in development and disease in eukaryotes and regulates phenotypes in prokaryotes. Aberrant methylation profiles that characterize diseases have potential for biomarkers and precise epigenomic therapies.
NEB offers a range of solutions for investigating DNA methylation. Enzymatic conversion is highly recommended over conventional bisulfite conversion to generate more accurate and sensitive 5mC results with next generation sequencing or PCR. The EpiMark® and NEBNext® kits simplify detection, amplification, and mapping of methylated or hydroxymethylated cytosine with enzymatic conversion modules, next generation sequencing library preparation kits with enzymatic conversion integrated, and validated primer sets for multiplexing. If conventional bisulfite conversion is desired, optimized DNA polymerases, high yielding library preparation kits, and targeted sequencing techniques are useful to address challenges for amplification and sequencing. Enrichment kits using high affinity MBD2-Fc fusion protein and magnetic bead capture of methylated CpGs from genomic DNA offer a cost-effective approach for genome-wide DNA methylation studies.
