    A suspension of E. coli strain ER2420 containing the plasmid pACYC177 which has been grown in LB medium and brought to 50% glycerol. ER2420 background: EcoK r- m- /McrBC- Mrr Dcm+.

    Growth Medium and Conditions

    The plasmid pACYC177 carries a gene encoding resistance to ampicillin (beta-lactamase) and a gene encoding resistance to kanamycin (kanamycin phosphotransferase). To maintain the plasmid cells should be grown with 50-100 µg/ml of amp and 35-50 µg/ml of kanamycin. The plasmid, which is present at a low copy number, should be amplified using a standard chloramphenicol procedure.
