    Description

    Blue Loading Buffer Reagents for SDS-PAGE:

    3X SDS Blue Loading Buffer (8 ml):
    187.5 mM Tris-HCl (pH 6.8 @ 25°C)
    6% (w/v) SDS
    30% glycerol and 0.03% (w/v) bromophenol blue
    (Store at room temperature)

    30X Reducing Agent (1 ml):
    1.25 M DTT
    (Store at -20°C)

    -20°C

    Notes

    1. Reagents stored at different temperatures.
    2. Prepare fresh 3X Reducing Blue Loading Buffer by adding 1/10 volume 30X Reducing Agent to 1 volume of 3X Blue Loading Buffer.
    3. Prepare samples by adding 2 volumes of Sample to 1 volume 3X Reducing Blue Loading Buffer (from step 1).
    4. Heat samples to 95-100°C for 3-5 minutes.
    5. After a quick microcentrifuge spin, load directly on to a gel. To ensure uniform mobiity, load an equal volume of 1X Reducing Blue Loading Buffer into any unused wells.
