FAQ: What are the differences between BsaI (#R0535), BsaI-HF (#R3535) and BsaI-HFv2 (#R3733)?
BsaI HFv2 has been optimized for Golden Gate Assembly but also works well for any protocol requiring DNA cutting by BsaI. The BsaI-HFv2 produces more efficient assembly and is not affected by sequence variation close to the recognition site that has been observed with some forms of BsaI. For any purpose requiring digestion at this recognition sequence (...5’-GGTCTC(N1)/(N5)-3’...) BsaI-HFv2 is the enzyme we recommend.
