    EcoP15I is a Type III restriction enzyme and therefore requires two copies of a non-palindromic recognition sequence, in inverse orientation, in the substrate DNA molecule to achieve efficient cleavage. Reading the top strand in the 5'-->3' direction, at some point you must have the following sequence…CAGCAGNCTGCTG…where "N" can be 0-12Kb in length.
