Fragmentation of the RNA will occur in the First Strand Buffer. If you are following the NEBNext Ultra Directional Workflow, we recommend addition of Actinomycin D to the First Strand Synthesis Reaction to increase strand-specificity. Actinomycin D is available from suppliers including Sigma (#A1410) and this should be dissolved in dimethylsulfoxide [DMSO] as described in the NEBNext product manuals.
