FAQ: Why does the RT primer hybridization occur before 5’adaptor ligation?
The small RNA library prep protocol has been improved to prevent adaptor-dimer formation. The RT primer is added to anneal with the un-ligated 3’ adaptor and transform a single-stranded DNA adaptor in to a double-stranded DNA molecule that is no longer a substrate for T4 RNA ligase 1.
