If your buffer is one of the following: 
Thermopol, stadard Taq, Phusion HF, Pusion GC, or DyNazyme Ext buffer then you can use the standard protocol. 

If your buffer is one of the following: 
AccuTaqLA. Pfu, iTaq, or Accu-prime Pfx then use the sequential reaction protocol.
