FAQ: Are there any special considerations when working with BbvI?
Like many Type IIS (shifted cut site) restriction enzymes, BbvI seems to remain bound to its substrate after cleavage. Extended digests are not recommended because 1 unit is required to cleave 1 μg of DNA in 16 hours. Phenol extraction is the suggested method of DNA purification due to the binding properties of the enzyme.
