    Yes, SacI-HF and SacI differ in their sensitivity to overlapping mammalian CpG methylation. SacI-HF™ is blocked by overlapping methylation on both strands and impaired if one strand is methylated. SacI cuts if one or both strands contain overlapping CpG methylation.
