FAQ: Will CIP dephosphorylate proteins?

CIP can be used to remove phosphates from phosphoserine/threonine and phosphotyrosine residues in proteins with a preference for phosphotyrosine residues. Typically 10-50 units of CIP will remove phosphate groups from 100-500 pmol of protein (5-25 mg of 50 kDa protein) in a 30 minute reaction under the recommended reaction conditions. Please note that optimal incubation time and enzyme concentration must be determined empirically for a particular substrate.

For optimal dephosphorylation of proteins, NEB recommends using Lambda Protein Phosphatase (Lambda PP), (NEB #P0753 ).
